These three strains form the gut-studied core of the V-IOTIC formula. Each carries a specific strain code — not just a species name — and has been studied together as a complex in a randomised, double-blind, placebo-controlled clinical trial. Their presence in V-IOTIC is traceable, intentional and evidence-led.

Many gut probiotics on the market list species names — Lactobacillus plantarum, Lactobacillus rhamnosus — without a strain code. A species name alone gives you no way to verify the research behind that specific strain, because thousands of strains exist within each species and they do not all behave the same way. PBS067, LRH020 and BL050 are specific, patented and studied. That is the difference.


How the Strains Work

Protecting the Gut From Within

In vitro research — published in Applied Microbiology and Biotechnology 2015 — studied the SmilinGut strains against three of the most common enteric pathogens. The results were assessed using an overlay assay measuring inhibition, with competitive and metabolite-mediated mechanisms both evaluated.

Escherichia coli

All three SmilinGut strains demonstrated strong inhibitory activity against E. coli — one of the most common causes of gut infection and digestive disruption.

Enterococcus faecalis

The strains showed significant competitive inhibition against E. faecalis, with synergic effects observed when strains were combined — demonstrating the value of a multi-strain approach.

Staphylococcus aureus

Strong acidic metabolite-mediated inhibition was observed against S. aureus, with the strains producing short-chain fatty acids and bacteriocins that compete with the pathogen.

The strains also demonstrated anti-inflammatory and antioxidant activity in vitro — showing significant effects on pro-inflammatory cytokines TNF-α and anti-inflammatory markers IL-4 and IL-10. This supports the idea that these strains do not just compete with pathogens — they actively support the gut's immune environment.


What It Addresses

The Gut Symptoms the Clinical Study Measured

The clinical study behind SmilinGut used a VAS Visual Analogue Scale symptom scoring system across five specific markers — all measured at regular intervals over 90 days including a 30-day washout period. These are the symptoms most commonly associated with gut microbiome disruption and IBS-C like presentations.

Bloating

Abdominal swelling and distension — one of the most common and disruptive gut symptoms, measured from baseline through the full study and washout period.

Flatulence

Excess gas and wind — assessed alongside bloating as part of the broader picture of gut fermentation and microbiome imbalance.

Abdominal Pain

General abdominal discomfort — a central symptom in IBS and gut dysbiosis, tracked consistently across the study timeline.

Abdominal Cramps

Cramping distinct from general pain — measured separately to capture the spasmodic discomfort associated with bowel irregularity.

Constipation

Infrequent or difficult bowel movements — a defining feature of IBS-C and a common consequence of gut microbiome disruption.


The Clinical Evidence

What the SmilinGut Study Reported

The clinical study was a randomised, double-blind, placebo-controlled trial of 50 subjects over 60 days, with stool collection, PCR analysis and VAS symptom questionnaires at T0, T10, T30, T60 and T90 including a 30-day washout. Published in BioMed Research International 2016, it is the gold standard in clinical research methodology.

Good results across all symptom indicators from day 10

The study reported meaningful improvements in bloating, flatulence, abdominal pain, abdominal cramps and constipation beginning at just 10 days — the first measurement point after baseline. All five symptom scores showed a downward trend in the active group from T10 onwards, with no significant change in the placebo group.

30%+ Improvement

Subjects self-reported over 30% improvement in symptoms

Using self-assessment scoring, subjects in the active group declared over 30% improvement in their symptoms — a clinically meaningful threshold used to define a positive treatment response in IBS research. This was a consistent finding across the study period.

3 Strains

Selective, consistent GI colonisation of all administered strains

PCR analysis of stool samples confirmed selective, consistent and significant colonisation of the gut for all three administered strains in the active group p<0.01. No significant increase was observed in the placebo group for any strain. This confirms that these specific strains survive and colonise the GI tract — not just pass through it.

90 Day effect

Long-term effect persisted after end of treatment

At T90 — 30 days after the supplementation period ended — the active group's symptom scores remained consistently lower than both the placebo group and their own baseline values p<0.001. This long-term effect after cessation of treatment is a strong indicator that the strains produced lasting changes in the gut environment, not just temporary symptom relief.

These findings relate to the SynbÆctive® SmilinGut strains studied under clinical conditions and published in peer-reviewed journals. Individual experiences can vary. V-IOTIC is a food supplement designed to support daily microbiome balance — it is not a medicine.

The Bigger Picture

Why Gut Health Belongs in a Vaginal-First Formula

The SmilinGut clinical study showed meaningful symptom improvements from as early as day 10, with the full study running over 60 days. Crucially, the long-term effect persisted 30 days after supplementation ended — which means the strains were not just providing temporary relief, but building lasting changes in the gut microbiome.

Consistency is everything. The strains need time to colonise, compete and establish themselves in the gut environment. A minimum of 10 days is required to see early movement, but the most meaningful and lasting outcomes in the clinical data appeared at 30, 60 and 90 days of consistent daily use.
