The Basics
What Is the Vaginal Microbiome?
The vaginal microbiome is the collection of microorganisms — primarily bacteria — that live in the vaginal environment. In a healthy state, it is dominated by Lactobacillus species, which produce lactic acid, maintain a low acidic pH, and create an environment that is naturally resistant to disruption.
pH Balance
A healthy vaginal pH sits between 3.8 and 4.5. Lactobacilli produce lactic acid to maintain this level, keeping unwanted organisms in check.
Natural Defence
A Lactobacillus-dominant microbiome acts as a first line of defence — competing for space and producing compounds that inhibit harmful bacteria and fungi.
It Changes With You
Hormones, antibiotics, diet, stress and life stage all shift the vaginal microbiome. It is not static — which is exactly why ongoing support matters.
When this balance is disrupted — when Lactobacilli levels fall and other bacteria move in — the result is vaginal dysbiosis. This can manifest as recurring discomfort, unusual odour or discharge, dryness, itching, or a persistent sense that something is simply not right. The Lead Strain
Why Lactobacillus crispatus Is the Strain That Matters Most
Of all the Lactobacillus species found in the vaginal environment, Lactobacillus crispatus is consistently associated with the healthiest vaginal microbiome profiles. It is the dominant species in women with low vaginal pH, minimal dysbiosis, and the fewest recurring intimate health issues.
It is also the strain most depleted during periods of imbalance — and the hardest to restore without targeted supplementation.
Many women's probiotics do list Lactobacillus crispatus on the label — but listing a species name is not the same as using a traceable, clinically studied strain. Without a strain code, there is no published research to reference, no way to verify what you are actually taking, and no evidence to link that specific strain to any meaningful outcome. A species name alone tells you very little about whether it works.

Lactobacillus crispatus LCR031 is the most prevalent species in a Lactobacillus-dominant vaginal microbiome and the most clinically researched strain in the context of vaginal health. It is the first strain selected for V-IOTIC, and the highest-dosed in the formula.
How the Strains Work
Competing With the Organisms That Cause Disruption
The SynbÆctive® Femme strains — which form the core of the V-IOTIC formula — have been studied in vitro for their ability to interact with three of the most common organisms associated with vaginal imbalance. Published in the International Journal of Molecular Sciences 2023, the research used electron microscopy at high magnification to observe what happens when the probiotic strains encounter these organisms directly.
Candida albicans
At 20,000x magnification, the Femme strains were observed forming a matrix around C. albicans cells, causing them to aggregate together and reducing their ability to adhere freely.
Escherichia coli
The Femme strains demonstrated direct interaction with E. coli cells — competing for surface attachment and limiting the organism's ability to establish itself.
Gardnerella vaginalis
The strains were able to form a biofilm that left no available surface for G. vaginalis to colonise — the organism most associated with bacterial vaginosis.
This is not a passive process. The Femme strains actively compete, adhere and form protective structures — which is why strain selection matters far more than CFU count alone.
The Clinical Evidence
What the Femme Studies Reported
The SynbÆctive® Femme complex has been studied in three published clinical trials covering vaginal colonisation, bacterial vaginosis and menopause. All findings below relate directly to the strains used in V-IOTIC — this is not generalised probiotic data.
Vaginal colonisation confirmed from oral intake
In a randomised, double-blind, placebo-controlled study of 20 women, all three Femme strains were detected in vaginal swabs after just 7 days of oral supplementation — confirmed by PCR analysis. Colonisation persisted through the 14-day treatment period and into the 7-day washout. The placebo group showed no significant change for any strain. Participants also reported a reduction in itching, burning, discharge and dryness in over 80% of cases.
Vaginal microbiome restoration
In a clinical study of 75 women, long-term supplementation with the Femme strains restored a Lactobacillus-dominant vaginal microbiome in over 90% of participants in the active group — with complete resolution of symptoms. In the control group, 30% of subjects still showed a partially or fully disrupted microbiome by the end of the study period. The data was published in Advances in Infectious Diseases 2019.
Reduction in recurrence within the study setting
The same 75-woman study monitored recurrence rates over six months. The control group showed a 40% recurrence rate. The active group — taking the Femme strains — showed a recurrence rate of 16% p<0.001. The active strains also reduced all clinical markers assessed during both the active phase and the maintenance period.
Menopause: vaginal health index improved by up to 50%
In a prospective observational clinical trial of 50 post-menopausal women published in Nutrients, 2024, the Femme strains demonstrated a significant reduction in pro-inflammatory markers including IL-6, TNF-α and IL-1β. Vaginal Health Index scores — assessed by gynaecologists evaluating elasticity, secretion and comfort — improved by up to 50% after four weeks of supplementation. Lactobacillus levels increased by 28%.
